Effects of Water Extract of Sibjeondaebo-tanggami on Cultured Spinal Sensory Neurons Damaged by Hydrogen Peroxide

Abstract

|
Objectives: This study was carried out to evaluate the effect of Sibjeondaebo-tanggami(SJDBTGM) water extract on cultured mouse spinal sensory neurons which was damaged by hydrogen peroxide.
Methods: The cultured mouse spinal sensory neurons was damaged by hydrogen peroxide-induced oxygen radicals. MTT<3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide> assay, NR(neutral red) assay, Neurofilament EIA(enzyme immuno assay) and TBARS assay were carried out after the cultured mouse spinal sensory neurons were preincubated with various concentrations of SJDBTGM water extract for 3 hours prior to exposure of hydrogen peroxide.
Results:
1. Hydrogen peroxide decreased the survival rate of the cultured mouse spinal sensory neuron cells on NR assay and MTT assay.
2. SJDBTGM have efficacy of increasing the amount of neurofilament decreased by hydrogen peroxide.
3. SJDBTGM have efficacy of decreasing the amount of lipid peroxidation increased by hydrogen peroxide.
Conclusions: From the above results, It is concluded that SJDBTGM has marked an efficacy as a treatment for the damages caused by hydrogen peroxide.
